    On-Demand Mobility (ODM) Technical Pathway: Enabling Ease of Use and Safety

    On-demand mobility (ODM) through aviation refers to the ability to quickly and easily move people or equivalent cargo without delays introduced by lack of, or infrequently, scheduled service. A necessary attribute of ODM is that it be easy to use, requiring a minimum of special training, skills, or workload. Fully-autonomous vehicles would provide the ultimate in ease-of-use (EU) but are currently unproven for safety-critical applications outside of a few, situationally constrained applications (e.g. automated trains operating in segregated systems). Applied to aviation, the current and near-future state of the art of full-autonomy, may entail undesirable trade-offs such as very conservative operational margins resulting in reduced trip reliability and transportation utility. Furthermore, acceptance by potential users and regulatory authorities will be challenging without confidence in autonomous systems in developed in less critical, but still challenging applications. A question for the aviation community is how we can best develop practical ease-of-use for aircraft that are sized to carry a small number of passengers (e.g. 1-9) or equivalent cargo. Such development is unlikely to be a single event, but rather a managed, evolutionary process where responsibility and authority transitions from human to automation agents as operational experience is gained with increasingly intelligent systems. This talk presents a technology road map being developed at NASA Langley, as part of an overall strategy to foster ODM, for the development of ease-of-use for ODM aviation

    The Naturalistic Flight Deck System: An Integrated System Concept for Improved Single-Pilot Operations

    This paper reviews current and emerging operational experiences, technologies, and human-machine interaction theories to develop an integrated flight system concept designed to increase the safety, reliability, and performance of single-pilot operations in an increasingly accommodating but stringent national airspace system. This concept, know as the Naturalistic Flight Deck (NFD), uses a form of human-centered automation known as complementary-automation (or complemation) to structure the relationship between the human operator and the aircraft as independent, collaborative agents having complimentary capabilities. The human provides commonsense knowledge, general intelligence, and creative thinking, while the machine contributes specialized intelligence and control, extreme vigilance, resistance to fatigue, and encyclopedic memory. To support the development of the NFD, an initial Concept of Operations has been created and selected normal and non-normal scenarios are presented in this document

    Anosmin-1 contributes to brain tumor malignancy through integrin signal pathways

    Anosmin-1, encoded by the KAL1 gene, is an extracellular matrix (ECM)-associated protein which plays essential roles in the establishment of olfactory and GNRH neurons during early brain development. Loss-of-function mutations of KAL1 results in Kallmann syndrome with delayed puberty and anosmia. There is, however, little comprehension of its role in the developed brain. As reactivation of developmental signal pathways often takes part in tumorigenesis, we investigated if anosmin-1-mediated cellular mechanisms associated with brain tumors. Our meta-analysis of gene expression profiles of patients' samples and public microarray datasets indicated that KAL1 mRNA was significantly upregulated in high-grade primary brain tumors compared with the normal brain and low-grade tumors. The tumor-promoting capacity of anosmin-1 was demonstrated in the glioblastoma cell lines, where anosmin-1 enhanced cell motility and proliferation. Notably, anosmin-1 formed a part of active β1 integrin complex, inducing downstream signaling pathways. ShRNA-mediated knockdown of anosmin-1 attenuated motility and growth of tumor cells and induced apoptosis. Anosmin-1 may also enhance the invasion of tumor cells within the ECM by modulating cell adhesion and activating extracellular proteases. In a mouse xenograft model, anosmin-1-expressing tumors grew faster, indicating the role of anosmin-1 in tumor microenvironment in vivo. Combined, these data suggest that anosmin-1 can facilitate tumor cell proliferation, migration, invasion, and survival. Therefore, although the normal function of anosmin-1 is required in the proper development of GNRH neurons, overexpression of anosmin-1 in the developed brain may be an underlying mechanism for some brain tumors

    Second Annual Transformative Vertical Flight Concepts Workshop: Enabling New Flight Concepts Through Novel Propulsion and Energy Architectures

    On August 3rd and 4th, 2015, a workshop was held at the NASA Ames Research Center, located at the Moffett Federal Airfield in California to explore the aviation communities interest in Transformative Vertical Flight (TVF) Concepts. The Workshop was sponsored by the AHS International (AHS), the American Institute of Aeronautics and Astronautics (AIAA), the National Aeronautics and Space Administration (NASA), and hosted by the NASA Aeronautics Research Institute (NARI). This second annual workshop built on the success and enthusiasm generated by the first TVF Workshop held in Washington, DC in August of 2014. The previous Workshop identified the existence of a multi-disciplinary community interested in this topic and established a consensus among the participants that opportunities to establish further collaborations in this area are warranted. The desire to conduct a series of annual workshops augmented by online virtual technical seminars to strengthen the TVF community and continue planning for advocacy and collaboration was a direct outcome of the first Workshop. The second Workshop organizers focused on four desired action-oriented outcomes. The first was to establish and document common stakeholder needs and areas of potential collaborations. This includes advocacy strategies to encourage the future success of unconventional vertiport capable flight concept solutions that are enabled by emerging technologies. The second was to assemble a community that can collaborate on new conceptual design and analysis tools to permit novel configuration paths with far greater multi-disciplinary coupling (i.e., aero-propulsive-control) to be investigated. The third was to establish a community to develop and deploy regulatory guidelines. This community would have the potential to initiate formation of an American Society for Testing and Materials (ASTM) F44 Committee Subgroup for the development of consensus-based certification standards for General Aviation scale vertiport capable flight systems. These standards need to accommodate novel fixed wing concepts that do not fit within the existing Federal Aviation Administration (FAA) rotorcraft certification framework (Code of Federal Regulations, Title 14, Chapter I, Subchapter C, Part 27). The fourth desired outcome was to launch an information campaign to ensure key U.S. Government agencies understand the potential benefits and industry interest in establishing new vertiport capable flight markets.     Upscaling Wetland Methane Emissions From the FLUXNET-CH4 Eddy Covariance Network (UpCH4 v1.0): Model Development, Network Assessment, and Budget Comparison

    Wetlands are responsible for 20%-31% of global methane (CH4) emissions and account for a large source of uncertainty in the global CH4 budget. Data-driven upscaling of CH4 fluxes from eddy covariance measurements can provide new and independent bottom-up estimates of wetland CH4 emissions. Here, we develop a six-predictor random forest upscaling model (UpCH4), trained on 119 site-years of eddy covariance CH4 flux data from 43 freshwater wetland sites in the FLUXNET-CH4 Community Product. Network patterns in site-level annual means and mean seasonal cycles of CH4 fluxes were reproduced accurately in tundra, boreal, and temperate regions (Nash-Sutcliffe Efficiency similar to 0.52-0.63 and 0.53). UpCH(4) estimated annual global wetland CH4 emissions of 146 +/- 43 TgCH4 y(-1) for 2001-2018 which agrees closely with current bottom-up land surface models (102-181 TgCH4 y(-1)) and overlaps with top-down atmospheric inversion models (155-200 TgCH4 y -1). However, UpCH4 diverged from both types of models in the spatial pattern and seasonal dynamics of tropical wetland emissions. We conclude that upscaling of eddy covariance CH4 fluxes has the potential to produce realistic extra-tropical wetland CH4 emissions estimates which will improve with more flux data. To reduce uncertainty in upscaled estimates, researchers could prioritize new wetland flux sites along humid-to-arid tropical climate gradients, from major rainforest basins (Congo, Amazon, and SE Asia), into monsoon (Bangladesh and India) and savannah regions (African Sahel) and be paired with improved knowledge of wetland extent seasonal dynamics in these regions. The monthly wetland methane products gridded at 0.25 degrees from UpCH4 are available via ORNL DAAC (https://doi.org/10.3334/ ORNLDAAC/2253).Plain Language Summary Wetlands account for a large share of global methane emissions to the atmosphere, but current estimates vary widely in magnitude (similar to 30% uncertainty on annual global emissions) and spatial distribution, with diverging predictions for tropical rice growing (e.g., Bengal basin), rainforest (e.g., Amazon basin), and floodplain savannah (e.g., Sudd) regions. Wetland methane model estimates could be improved by increased use of land surface methane flux data. Upscaling approaches use flux data collected across globally distributed measurement networks in a machine learning framework to extrapolate fluxes in space and time. Here, we train and evaluate a methane upscaling model (UpCH4) and use it to generate monthly, globally gridded wetland methane emissions estimates for 2001-2018. The UpCH4 model uses only six predictor variables among which temperature is dominant. Global annual methane emissions estimates and associated uncertainty ranges from upscaling fall within state-of-the-art model ensemble estimates from the Global Carbon Project (GCP) methane budget. In some tropical regions, the spatial pattern of UpCH4 emissions diverged from GCP predictions, however, inclusion of flux measurements from additional ground-based sites, together with refined maps of tropical wetlands extent, could reduce these prediction uncertainties

    The Overseeing Mother: Revisiting the Frontal-Pose Lady in the Wu Family Shrines in Second Century China

    Located in present-day Jiaxiang in Shandong province, the Wu family shrines built during the second century in the Eastern Han dynasty (25–220) were among the best-known works in Chinese art history. Although for centuries scholars have exhaustively studied the pictorial programs, the frontal-pose female image situated on the second floor of the central pavilion carved at the rear wall of the shrines has remained a question. Beginning with the woman’s eyes, this article demonstrates that the image is more than a generic portrait (“hard motif ”), but rather represents “feminine overseeing from above” (“soft motif ”). This synthetic motif combines three different earlier motifs – the frontal-pose hostess enjoying entertainment, the elevated spectator, and the Queen Mother of the West. By creatively fusing the three motifs into one unity, the Jiaxiang artists lent to the frontal-pose lady a unique power: she not only dominated the center of the composition, but also, like a divine being, commanded a unified view of the surroundings on the lofty building, hence echoing the political reality of the empress mother’s “overseeing the court” in the second century during Eastern Han dynasty

    Basic science232. Certolizumab pegol prevents pro-inflammatory alterations in endothelial cell function

    Background: Cardiovascular disease is a major comorbidity of rheumatoid arthritis (RA) and a leading cause of death. Chronic systemic inflammation involving tumour necrosis factor alpha (TNF) could contribute to endothelial activation and atherogenesis. A number of anti-TNF therapies are in current use for the treatment of RA, including certolizumab pegol (CZP), (Cimzia ®; UCB, Belgium). Anti-TNF therapy has been associated with reduced clinical cardiovascular disease risk and ameliorated vascular function in RA patients. However, the specific effects of TNF inhibitors on endothelial cell function are largely unknown. Our aim was to investigate the mechanisms underpinning CZP effects on TNF-activated human endothelial cells. Methods: Human aortic endothelial cells (HAoECs) were cultured in vitro and exposed to a) TNF alone, b) TNF plus CZP, or c) neither agent. Microarray analysis was used to examine the transcriptional profile of cells treated for 6 hrs and quantitative polymerase chain reaction (qPCR) analysed gene expression at 1, 3, 6 and 24 hrs. NF-κB localization and IκB degradation were investigated using immunocytochemistry, high content analysis and western blotting. Flow cytometry was conducted to detect microparticle release from HAoECs. Results: Transcriptional profiling revealed that while TNF alone had strong effects on endothelial gene expression, TNF and CZP in combination produced a global gene expression pattern similar to untreated control. The two most highly up-regulated genes in response to TNF treatment were adhesion molecules E-selectin and VCAM-1 (q 0.2 compared to control; p > 0.05 compared to TNF alone). The NF-κB pathway was confirmed as a downstream target of TNF-induced HAoEC activation, via nuclear translocation of NF-κB and degradation of IκB, effects which were abolished by treatment with CZP. In addition, flow cytometry detected an increased production of endothelial microparticles in TNF-activated HAoECs, which was prevented by treatment with CZP. Conclusions: We have found at a cellular level that a clinically available TNF inhibitor, CZP reduces the expression of adhesion molecule expression, and prevents TNF-induced activation of the NF-κB pathway. Furthermore, CZP prevents the production of microparticles by activated endothelial cells. This could be central to the prevention of inflammatory environments underlying these conditions and measurement of microparticles has potential as a novel prognostic marker for future cardiovascular events in this patient group.
